The vision of the SINPAIN is to improve the quality of life for those affected by OA and reduce the substantial costs linked with the disease by developing a new generation of OA therapeutics.

Innovative nanotherapeutics pipeline

Develop an innovative nanotherapeutics pipeline based on siRNA for treating different stages of knee OA, early (grade 0-1) and later stages (grade 3-4)

New understanding of the complex disease

Add to a new understanding of the complex disease, its underlying causes and mechanisms, allowing for more personalised diagnostic and treatment options for knee OA

Technology

Obtain technology that will be instrumental in alleviating pain and slowing down disease progression for countless patients

RESEARCH AREA

Osteoarthritis (OA) is one major course of years lived with disability in the elderly and considered a high burden disease. There is no cure for OA and available treatments needs to be reconsidered. Current pharmacological interventions consist mainly of analgesic and anti-inflammatory drugs with the addition of intraarticular steroids and hyaluronic acid (IA- HA) injections.

Nevertheless, all with moderate efficacy and associated long-term side effects. Taking advantage of the explosion of RNA technologies in the last years, SINPAIN aims to develop a pipeline of siRNA-based therapy built on the combination of current technologies (dynamic IA-HA and nanocarriers) that will be designed step- by-step in order to reach a successful management of inflammation and innervation therapy for the treatment of early (grade 0-1) and later stages (grade 3-4) of knee OA. Exploiting the data obtained during the project, a decision-making tool based on machine learning will be validated to offer patients a personalized therapy.

Research

So far, osteoarthritis (OA) has no cure, and current treatment approaches suffer many drawbacks. Moreover, the number of people affected by the progressive disease is steadily rising, entailing a growing unmet need for therapeutic OA treatments.

SINPAIN strives to become a game-changer in knee osteoarthritis (OA) treatment by developing a product pipeline for safe, efficient, and cost-effective advanced siRNA therapies.

Know-how

The proposed SINPAIN therapy will combine RNA technology, nanocarriers and an improved hyaluronic acid-based knee visco-supplement (IA-HA). The pioneering treatment has the potential to manage inflammation and control the innervation of knee OA at any stage of the disease progression (grades 0-1 and 3-4). Importantly, SINPAIN will study the molecular pathways underpinning OA by state-of-the-art characterisation techniques to discover interplaying mechanisms. 

Specifically designed machine-learning tools will support the delivery of personalised therapies.

THE ROLE OF UPO

In the EU research project UPO is leading imaging mass cytometry and single cell transcriptomic/proteomic research on the biopsies of the OA affected individuals. UPO is custom-designing and validating imaging mass cytometry panels in order to precisely identify blood vessels, synovial cells, cartilage, nerve fibbers, pericytes, T and B-cell, and macrophages. 

UPO is also participating in the assessing of efficacy of the clinically relevant in vivo OA model and in demonstrating in vivo biocompatibility of the advanced nanotherapeutic cell.
